Chronic myelomonocytic leukaemia (CMML) is a clonal haematopoietic stem cell malignancy with few therapeutic options and dismal prognosis. It is characterised by epigenetic and transcriptional dysregulation, exemplified by the presence of recurrent mutations in epigenetic and splicing factors in >90% and >70% of cases, respectively. The successful candidate will investigate the mechanistic basis of aberrant splicing in CMML and its interaction with epigenetic dysregulation in driving disease initiation and progression. They will have access to state-of-the-art technology and an extensive Biobank of primary human material, both normal and leukaemic. Work will include generation of in vitro and in vivo models and generation/integration of transcriptomic, epigenomic and proteogenomic datasets. Downstream targets of mechanistic relevance will be validated and explored as potential therapeutic targets. They will also have the opportunity to test novel spliceosome inhibitors in appropriate model systems. Dr Wiseman’s group is located within the state-of-the-art Manchester Cancer Research Centre (MCRC), and is highly integrated with a tertiary referral clinical practice based at the adjacent Christie Hospital.

 

Dr. Wiseman’s group is co-led by Dr Kiran Batta, and works closely with Prof. Tim Somervaille’s Leukaemia Biology Group, thereby providing a stimulating collaborative environment with access to a diverse range of expertise and resources. There are numerous active collaborations with leaders in the field of CMML biology, both nationally and internationally. This post is funded by Dr Wiseman’s Oglesby Leukaemia Research Fellowship.

 

You will be expected to hold a PhD (or equivalent) qualification in a relevant field with appropriate specialist research experience in molecular biology and cell biology. Desirable areas of experience would include normal and malignant haematopoiesis, transcriptional regulation, splicing, epigenetics, next generation sequencing, flow cytometry and advanced data analysis are desirable.
